Cmu inset

Desired building size of 30’x16’. Monolithic slab with 1 course of cmu’s.

Is it advisable to make the slab an inch or two larger and inset the block that inch or two so that the cmu are not set at the edge of the slab )in this case footer?

I wouldn't inset them. You're only creating a place for rain, snow, ice, dirt and debris to sit.
