Just pouring slab. I plan a future mini split install. Should I make any penetrations through my slab in preparation for the mini split install?

The typical mini-split installation run straight thru an exterior wall, above grade. It you want to "bury" the pipes in the wall, you need a 2x6 (or even 2x8) wall.

I ran mine through the walls, I wish I didn't. Just makes it difficult if you have to fix it.

Also had no issues running it through 2x4 framing

No problems running lines in a 2x4 wall.

However as it was mentionned it makes fixing things much more complicated.

It's not any more likely that you'll need to fix HVAC lines in your wall than having to fix plumbing or electrical in your wall. Pretty much a non-issue.

Probably more likely you'll damage it if it's not in the wall.
