If I use a HVAC Gaspack unit with heating and air or a Heat pump for one large area **** at 1000sqf can I have one big vent entering the building and one big return?

yes. just run the suppy all the way to the end and let the return air draw it back.

that will probably short cycle the unit. by looking at the pic it looks like 40x20? I would run 2 supply ducts down the wall one on each side with 2-3 diffusers on each. no need to insulate the duct if its an open space.

What kind of diffusers do I need?

It depends on CFM, duct type and location. For your application, you'll be concerned with ones for ceilings or overhead, or for vertical mounted ones, like on a wall. It depends on how you plan on laying out the supply duct.
