Typically there's enough safety factor in wood construction to deal with a reasonable amount of defects. I'm going to WAG its based on the #2 & Better standard of SPF and was probably worked out a long time ago.
It bothers me also but there's not much to do other than sister on another timber a couple feet in either direction, apply construction adhesive and nail off liberally.

Mine have the same issue. It bothered me as well. The builder said it was no big deal.

Graded lumber is graded based on species, grain, and knots- location and condition.
The strength of a piece of graded lumber is assured to meet some minimum based on all those possible 'conditions'
So if a builder (or their engineer) is doing their job, they have determiend that 'this post can use XYZ lumber and will hold ABC loads'\
So if your building was properly engineered, and the lumber accurately graded, and everyone did their job- it is of zero concern.
MY concern is not the knot falling out or staying in- MY concern would be 'did they use the proper lumber'. Filling the knots in will do jack squat.

I am not a structural engineer so my opinion isn't worth very much. All I will say is, I have seen other knots like that missing in other buildings so it must be fairly commonplace. Filling in the knots may make you feel better, and it will no doubt look better, but I doubt it would do anything in-so-far as the actual structural strength is concerned. But that is just an opinion from a non-engineer.

Thank you for your inquiry about the naughty wood and the multitude of pictures showing the same from the recent build of our company's pole barn for you.
While we can assure you that the naughty wood is by no means comprised in strength or performance we may not have been a diligent in telling you about our non naughty wood packages. You may recall them being referred to as our Artisanal Options 1,2 and 3.
Option 3 would have given you no obvious naughtiness but you may have been bothered by what you couldn't see so perhaps Artisanal Option 2 would have been better.
Option 2 comes with a certificate stating that no hidden naughtiness was noted and that only lumber that was of good character was used in the construction of your pole barn. But given the number of photos you sent us along with the documentation of each naughty perhaps Option 1 would have been best.
Option 1 allows for no naughtiness in anyway shape or form. Trees used have their DNA tested and each piece is CAT scanned to rule out even the hint of naughtiness. Coming from Old Growth Timbers that have been sourced with due regard to every environmental and ethical concern including asking the tree if it was ready to die and then given the appropriate counseling and cooling off period. With Option One even the most concerned buyer can rest easy.
You may have felt the 30,000 percent increase in price for Option one over the price of your Naughty Wood Package was out of reach but who can put a price on peace of mind. Certainly not someone with OCD!

Wont mean a thing, its partially overcome or fixed due to the fact there is 3 of them and in a column.
