I am building a new shop with ICF. 100Lx60Wx16H. Using 5” of open cell on backside of roof. Insulation under slab , 8” concrete.

I would like to use a single 10 ton unit with air handler in the attic and using a soft duct right down the center of the shop. Also going to install a big *** fan 15’ at the center point of shop.

Does anyone have suggestions for something that would work better

No windows in shop 3 insulated garage doors and 2 walk through doors.

That's a lot of insulation, but that's also a lot of shop. I would think you're going to need to distribute that air throughout the space more effectively than dumping it straight down the middle of the shop. If you were to do that you'd have to have an enormous output duct anyway. I would probably look at rigid tubing distributing the air throughout the space in a star shape - anything to help distribute the air better.
