Getting my "stuff" in order to do a 32'x56' pole barn, 4" concrete floor. What determines the size of Pex tubing that I need for future heat? As always, thanks.

Contact a radiant heat consultant to find out your BTU needs.

I used 1/2 pex about 12" apart in my similar sized floor space. BUT, I have 1 door. If you add windows, higher/lower etc it all changes. Contact a designer.

most of the companies that sell pex and heating supplies will figure it all out for for free. They did for me.

The size of pex is determined by how long of loops you run. 1/2 pex is good for 300' or less. With larger sizes you can go longer. Doing a heat loss on the building will determine spacing of the the loops.
