if you mean to inject a tone onto a pair of wires for tracing, I don't see why not (unless the pair in question are open or not going to the same termination). But the "other end" must not have a load that will drag the level down.

Alternate method :
1. disconnect wires at back of receiver
2. twist bare leads together of 1 wire set
3. on speaker end - with continuity tester (ohmmeter) ring out each lead until find shorted wire set
4. label each wire end at receiver
5. untwist set that of wires
6 repeat back to step 2

or
If speakers are already wired on speaker end. Disconnect all at receiver and 1 by 1 connect, see which speaker gives sound. Then label wire at receiver, then repeat.
