Stopped in at a garage sale and found a set of Spellmaco 28 pc. transfer punch set for $5. I have never used these kind of punches, what is the proper method for using these?

Are you describing centering transfer punches? If so you should just tap the punch lightly when it is in the hole so you make a slight mark on the new surface. Then use a normal punch to make the mark deeper to accept a drill tip.
The transfer punches will last much longer using this method.

If you have two 3/8'' holes in a plate that is 2'' x 4'' and want the holes matching on another 2'' x 4'' plate ; line up plates and use a 3/8'' transfer to mark the exact centers of the original. Tap lightly and the use a center punch to make the real center punch mark.

Well worth having in the arsenal. i have a cheap set and use them mainly on snapped off exhaust manifold studs in Honda small engines.

Thanks for the tip on just making a small mark. Its what I did instinctively as I have some experience, but as a young en, I would have smacked it good.

Please elaborate. How do you set up a drill press chuck to fly straight on a drill press using a transfer punch.

Unlike a twist drill bit, a smooth sided transfer punch can be used to check chuck wobble (runout) by sight, or more precisely with a dial indicator. I like to use 5/16 or 3/8. Set the quill first, then mount the transfer punch like a bit. You can see big problems right away. If you have over 2 thousands, knock the chuck out, remove the punch, roll the jaws in, and remount.

Use a small square against the chucked punch to square the table.

There are some good videos on YouTube.
