From the service to the building I have 2/0-2/0-2/0-1 URD in buried conduit. Now I need to get 100' feet into the building to the elec panel. If just use 2/0-2/0-2/0-1 SER without conduit, what do I need to do for protection? Just common sense? It will be in a steel barn.

Yes for SER u need to make sure its not subject to damage or in an area that it could get damaged.

How long is the whole run panel to panel?

What size breaker are u feeding this with?

About 450' run. Of course I came out 20' short of getting to the building :(

What do u plan on running on this feed? AC, heat, air compressor, any other large loads etc etc?

Unfortunately, due to the length u will have over 3% voltage drop even with a 50a load.

Is your building 100' long? If thats the case, u can expect even more voltage drop on the 120v branch circuits and Im guessing it will be over the recommended max 5% for feeder and branch circuit combined. If your voltage is already low at the main service panel, then u may have significant voltage issues at the building.
